Abstract/Description

The International Potato Center has developed several methods for detecting and identifying virus, but most methods require complicated and big equipment (i.e., thermal cycler), laboratory conditions and sensitive reagents that need a cold chain. Loop-mediated Isothermal amplification (LAMP), a newly developed nucleic acid amplification method, combines rapidity, simplicity, and high specificity in detecting pathogens. These features, combined with portable and battery-operated instruments and reagents tolerant to plant inhibitors, enable LAMP tests to be performed in farmer fields. We have developed two videos showing step by step the LAMP process for detecting sweetpotato viruses in both laboratory and field facilities. LAMP can be used in seed testing or surveillance testing schemes and considering that LAMP test can be rapidly adopted to detect different pathogens in different plants by just replacing some reagents (i.e., the specific primers for those pathogens) in the reaction is broadly applicable and adjustable to new pathogens.
